Folsäure ist ein B-Vitamin; in Lebensmitteln kommt es als Folat vor. In der Zelle trägt es als Tetrahydrofolat einzelne Kohlenstoff-Einheiten – für die Bausteine von DNA und RNA und für den Methionin-Kreislauf.

- Folat in der Nahrung → Folat-Monoglutamat Folat-Hydrolase Ein Enzym am Bürstensaum des Dünndarms spaltet die Glutamat-Kette ab, bis ein einziger Baustein übrig bleibt. Erst diese Form kann die Darmzelle aufnehmen. Quelle 1

- THF → 5,10-Methylen-THF SHMT · Serin, Vitamin B6 (PLP) Die SHMT überträgt eine Kohlenstoff-Einheit von Serin auf THF; ihr Cofaktor ist Vitamin B6. Aus dem Ergebnis stellt das Enzym MTHFR mit FAD wieder 5-Methyl-THF her, und der Kreis schließt sich. Quelle 2
- 5,10-Methylen-THF → dTMP Thymidylat-Synthase · dUMP Die Thymidylat-Synthase überträgt die Kohlenstoff-Einheit auf dUMP. So entsteht dTMP, einer der vier Bausteine der DNA. Quelle 2
